Biotic parameters in Shahid Rajaee reservoir (Iran)

Human activities in riverine water bodies can cause loading of nutrients and other pollutants in receiving systems e.g. reservoirs, affecting species interactions, population dynamics and ecosystem functioning. The present study aimed to investigate the influence of anthropogenic alteration on an artificial water body, i.e. the Shahid Rajaee reservoir (Mazandaran province, Northern Iran). The reservoir is subject to high anthropogenic nutrient loads from the Shirinroud and Sefidroud rivers, the effect of which needs to be evaluated considering the strategic importance of the reservoir as a source of water for the human population. A field sampling campaign was carried out at seven stations (one in each of the two river mouths and five in the reservoir itself) each month from April to July 2016. The study entailed sampling of benthic macroinvertebrate fauna at the river mouths and plankton communities at the reservoir stations. In addition to macroinvertebrate and plankton collections, physicochemical and nutrient variables were measured at all stations. Substantial loading of organic matter, mainly from the Shirinroud river, was detected throughout the sampling period, with a detrimental effect on both benthic and plankton communities. The decline in plankton diversity was particularly noticeable at stations close to the river mouths during June and July, with a bloom of Cyanobacteria and Dinoflagellates and strong dominance of larger zooplankton taxa, i.e. Cladocera. Our results suggest that river water quality, affected by human activities, alters the planktonic communities in Rajaee reservoir. Our assessment indicates that measures on both the Shirinroud river and the reservoir are required in order to mitigate and prevent even stronger eutrophication of the reservoir ecosystem.

Description
The study entailed sampling of benthic macroinvertebrate fauna at the river mouths and plankton communities at the reservoir stations. For phytoplankton, at each station three samples were taken, within 10 m distance. One litre of sub-surface water was collected using a Ruttner sampler (HYDRO-BIOS). The samples were immediately preserved in opaque containers in situ with Lugol’s iodine solution and transferred to the university of SANRU. Following Utermöhl (1958), a 50 ml sub-sample was poured into a settling chamber and left to settle for 24 h. The base plate was then removed from the chamber and a slide placed over the top of the base plate into which the cells had settled. The composition of the samples was analysed using an optical microscope (400× magnification), and the results were recorded as cells. Detailed taxonomic identification was performed with reference to Prescott (1954); Botes (2001); Wehr (2011); Josef (2014); Moestrup (1988). Zooplankton samples were collected using a plankton net (0.20 m2 mouth area, 50 µm mesh) towed along a 12-metre transect below the water surface. The samples then were preserved in Lugol’s iodine solution and transferred to the university of SANRU. Following (Heath 2001), the zooplankton specimens were enumerated and identified down to genus level under a microscope (100-400x magnification) using the keys provided in (Bruno et al. 2005; Defaye 1988; De Bernardi and Peters 1987; Voigt and Koste 1987). Specimen counts were performed in a Sedgwick-Rafter chamber and densities were expressed as individuals per litre. Benthic macroinvertebrates were sampled using a 60 µm-mesh Surber net with a surface area of 0.09 m2. At each station three samples were collected, one near each bank and one in the centre of the river. The samples were preserved in 75% alcohol and transferred to SANRU. The samples were sorted under a dissecting microscope and identified to the lowest possible taxonomic level, with reference to selected keys (Pescador et al. 2004; Elliott et al. 1988; Milligan 1997).

Sampling

Sampling Description
Data were collected in Shahid Rajaee reservoir located in the northern part of Iran. It was created by damming the Shirinroud and Sefidroud permanent tributaries and drains via the Tajan river to the Caspian Sea, with an annual average flow of 19.4 cubic meters/seconds. The average annual pre-dam discharge of Shirinroud and Sefidroud rivers jointly is 10.5 cubic meters/seconds. The surface area of the reservoir is 428 hectares at normal water level i.e., the level evidenced by the long-term presence of surface water as indicated directly by hydrophytic plants or hydric soils (Vuglinskiy 2009), and the total reservoir volume is 191.5×106 cubic meters. The area surrounding the reservoir is covered mainly by forest, natural rangeland, cropland and trout farms. The reservoir is used for flood control, and supplies water for hydroelectric power production and (more recently) drinking water for a population of 400 ×103 people located in Sari City (Makhlough et al. 2017). The sampling design was as follows: for phyplankton 4 times, 5 sampling stations and three samples; for Zooplankton 4 times, 5 sampling stations and 1 sample; for benthos 4 times, 2 sampling stations and three samples.
