          <para>Sub-samples for phytoplankton taxonomic identification and cell size / abundance analysis were preserved with Lugol (15 ml/l of sample). Samples were observed with a Nikon T300E inverted microscope following Utermöhl’s method and identified to species level where possible. Data were obtained and linear dimensions were measured at 400x magnification with a microscope connected to a video-interactive image analysis system (L.U.C.I.A, Version 4.8, Laboratory Imaging s.r.o.). Then, the calculation of volume was based on geometric approximations assigning a geometric shape for determination of surface area and volume following (https://www.phytovre.lifewatchitaly.eu/vre/shapes-groups/). As a general rule, traits were calculated for counting units, most often this means single cells and sometimes 100 ?m threads, coenobiums and colonies (e.g. in some filamentous cyanobacteria) where cells are not observable.</para>

Water samples were collected with horizontal tows from the subsurface (0.5 m) with a 6 μm plankton net. Water samples were fixed with Lugol’s solution.</para>

      <title>POR PUGLIA 2000/06 CIP PS_126 - Innovative methodological tools for the assessment of health status of aquatic ecosystems by phytoplankton microscopy analysis: eco-technologies and information technologies.</title>
      <abstract>
        <para>The project aimed to develop products with high technological content for classifying and monitoring the state of health of aquatic transitional and marine-coastal ecosystems, combining innovation in eco-technologies with innovation in information technologies. The purpose of the project was to develop a certification protocol for monitoring the state of health of aquatic coastal ecosystems, based on the analysis of the characteristics of the dimensional structure (a rough descriptor of the bodily dimensions of the individuals and the relative abundance of individuals of different bodily dimensions) of phytoplankton corporations, which, in terms of information content, technological potential and simplicity of use, are increasingly becoming the object of research all over the world.</para>
