The InBIO Barcoding Initiative Database: Lacewings (Neuroptera) 01

The dataset contains 234 records of Lacewings (Neuroptera) species collected from 2006 to 2019 in continental Portugal (Oliveira et al., 2021). Specimens were detected and captured by direct search of the environment and by using both UV and mercury-vapor lamps to attract the insects. Captured specimens were preserved in 96% ethanol. All captured specimens were identified to species level. Samples of each species were selected for DNA sequencing based on their geographic provenance. From each specimen, one leg (tissue sample) was removed to be used for DNA extraction. The DNA barcoding of these specimens was conducted within the InBIO Barcoding Initiative (IBI), funded by EnvMetaGen and PORBIOTA projects. DNA barcode sequences were deposited in BOLD (Barcode of Life Data System) and GenBank databases. All specimens and DNA extracts are deposited in the IBI collection at the CIBIO (Research Center in Biodiversity and Genetic Resources).

Abstract

The InBIO Barcoding Initiative aims to develop a reference collection of DNA barcoding sequences covering Portuguese invertebrate taxa. Within the project, a special focus is afforded to insects, given their relevance to food webs, ecosystems functioning, high diversity and limited number of DNA barcoding sequences of interest.

Funding

InBIO Barcoding Initiative is funded by the European Union’s Horizon 2020 Research and Innovation programme under grant agreement No 668981 and by the project PORBIOTA — Portuguese E-Infrastructure for Information and Research on Biodiversity (POCI-01-0145-FEDER-022127), supported by Operational Thematic Program for Competitiveness and Internationalization (POCI), under the PORTUGAL 2020 Partnership Agreement, through the European Regional Development Fund (FEDER). The fieldwork benefited from EDP Biodiversity Chair, the project “Promoção dos serviços de ecossistemas no Parque Natural Regional do Vale do Tua: Controlo de Pragas Agrícolas e Florestais por Morcegos” funded by the Agência de Desenvolvimento Regional do Vale do Tua, and includes research conducted at the Long Term Research Site of Baixo Sabor (LTER_EU_PT_002).

Study Area Description

Descriptor

Specimens were collected in 12 continental Portugal administrative districts. Eighty-seven different localities were sampled. Sampling was conducted from 2006 to 2019, although most specimens (87.6%) were collected in the period between 2017 to 2019.

Design Description

Description

The specimens in this dataset were collected in fieldwork incursions carried out between 2006 and 2019, encompassing several sites throughout continental Portugal in an attempt to have a good representation of the country’s Neuroptera fauna. Specimens were morphologically identified and DNA barcoded.

Methods

• Method

Method Step

Description

Specimens were collected in 87 different localities of continental Portugal. Fieldwork was carried out between 2006 and 2019, with 87.6% of the records obtained between 2017 and 2019.

Method Step

Description

Specimens were collected by direct search of the sampling site while also using both UV LED and mercury-vapor lamps as lures distributed along the site. Specimens were preserved in 96% alcohol and deposited in the IBI reference collection.

Method Step

Description

Specimens were morphologically identified using a stereoscopic microscope, an Olympus SZX2-ILLT, and resorting to most recent literature on Iberian Neuroptera (Monserrat, 2014a, 2014b, 2014c, 2016a, 2016b; Monserrat and Acevedo, 2013; Monserrat et al., 2012a; 2012b, 2014)

Method Step

Description

All specimens were DNA barcoded. For this, one leg was removed from each individual, DNA was then extracted, and a 658 bp COI DNA barcode fragment was amplified and sequenced. All tissue samples and DNA extracts were deposited in the IBI collection.

Method Step

Description

All sequences in the dataset were submitted to BOLD and GenBank databases and to each sequenced specimen the morphological identification was contrasted with the results of the BLAST of the newly generated DNA barcodes in BOLD Identification Engine.

Method Step

Description

Prior to GBIF submission, data was checked for errors and inconsistencies with OpenRefine 3.4 ( http://openrefine.org/).

Study Extent

Specimens were collected in 87 different localities of continental Portugal. Sampling was conducted from 2006 to 2019, in several fieldwork expeditions. Most specimens (87.6%) were collected in the period between 2017 to 2019.

Specimens were captured by direct search of the environment and by using both UV LED and mercury-vapor lamps to attract the insects. Specimens were preserved in 96% ethanol for downstream molecular analysis and stored at the InBIO Barcoding Initiative (IBI )reference collection.
