Description

Phytoplankton analysis was carried out using an inverted microscope (Nikon T300E) at 400 x magnification following Utermohl's method. Phytoplankton counts and morphometric measurements were performed semi-automatically using a video-interactive image analysis system (L.U.C.I.A Version 4.71, Laboratory Imaging) connected to the microscope. Using this system, the field of the Utermöhl's chamber observed through the microscope is reflected on the computer screen and every organism is associated with the most similar geometric shape selected from the set of proposed models. Given that the image is reflected on the computer screen, small organisms (C3 lm) are visible at 400 x magnification. By outlining the contour of the organism (single cell, colony) on the screen, morphometric information including length, width and greatest axial linear dimension (GALD lm) was obtained directly. From these measurements, the organism biovolume (V, cubic micrpmeter) (hereafter organism size), surface area (S, square micrometer) and surface-to-volume ratio were calculated . A geometric shape identified with a numerical code was attributed to each taxon.

Sampling

Sampling Description

Water samples were collected during four oceanographic out in March, June, September and December 2000 along the Southern coast of Puglia (Adriatic and Ionian Seas, SE Italy). Water phytoplankton samples were collected using a Niskin bottle at depths of 0.5 m at 21 sampling sites distributed along 7 transects running perpendicular to the coastline, with 3 sampling sites per transect located at 3, 9 and 15 nautical miles from the shore. Phytoplankton samples were preserved in situ with Lugol's iodine solution (15 ml per litre of sample). Samples were observed with a Nikon T300E inverted microscope connected to a video-interactive image analysis system (L.U.C.I.A, Version 4.71, Laboratory Imaging s.r.o.).
